近日,來自湖南大學(xué)化學(xué)生物傳感與計(jì)量學(xué)國家重點(diǎn)實(shí)驗(yàn)室的聶舟教授課題組在細(xì)胞調(diào)控領(lǐng)域取得重要進(jìn)展,在國際頂級期刊《Angew. Chem. Int. Ed.》(IF=12.102)在線發(fā)表研究論文《A DNA-mediated Chemically Induced Dimerization (D-CID) Nanodevice for Nongenetic Engineering of Receptor to Control Cell Behavior》,并被該期刊選為“Hot Paper”。


研究成果

細(xì)胞行為的精確控制是目前細(xì)胞治療技術(shù)中的難點(diǎn)和熱點(diǎn)問題。


聶舟教授課題組開發(fā)了一種基于DNA納米機(jī)器的受體非遺傳改造策略,對細(xì)胞膜表面受體特異性進(jìn)行定制化改造,通過DNA納米技術(shù)賦予其化學(xué)小分子響應(yīng)性,實(shí)現(xiàn)了小分子調(diào)控的細(xì)胞行為。


該策略應(yīng)用DNA介導(dǎo)化學(xué)誘導(dǎo)二聚(D-CID)實(shí)現(xiàn)了受體酪氨酸激酶的二聚激活,并介導(dǎo)下游的細(xì)胞信號傳導(dǎo)途徑,可對細(xì)胞遷移行為進(jìn)行精確控制,無需復(fù)雜遺傳改造即可賦予細(xì)胞特定的化學(xué)趨向性。進(jìn)一步通過不同的DNA功能模塊組合,并利用微流控裝置構(gòu)建復(fù)雜多細(xì)胞環(huán)境,成功證明了該策略可用于多目標(biāo)細(xì)胞行為的多化學(xué)因子精準(zhǔn)正交控制,有望實(shí)現(xiàn)在復(fù)雜細(xì)胞環(huán)境下對特定靶細(xì)胞類型的智能行為調(diào)控。


D-CID策略由于其DNA識別單元的多樣性,因此具有小分子配體及對應(yīng)細(xì)胞膜受體的普適性。

這種細(xì)胞行為的智能化調(diào)控策略有望為細(xì)胞治療及再生醫(yī)學(xué)的體內(nèi)應(yīng)用提供一種新思路。
